Set up the assay as follows:

1) Make doubling dilutions of the two serum samples as shown (the samples you have been given are already diluted 1:100).
2) The wells of the plate have already been coated with CMV antigen.

3) Add 200µl of serum to each well in the pattern shown opposite.
Use a fresh pipette tip for each serum, starting from the highest dilution (i.e. lowest concentration) and working towards the lowest dilution.

4) Add 200µl of undiluted serum to the CMV-negative and CMV-positive control wells (columns 9 and 10).

5) Add 200µl of the diluent to the diluent control wells (column 11).

Do not add anything to the wells in column 1 and 12 yet.

Incubate the plate in a humidified box for 1 hour at 37°C to allow any antibody present to attach to the antigen-coated wells
