Haemagglutination Inhibition Assay:

  1. Put 50µl of PBS into the following wells of the microtitre plate:
  2. C1-C10
  3. D1-D10
  4. E1-E10
  5. Put 50µl of antiserum in the first well of each row (use one row for each antiserum). Mix and transfer 50µl to second well, and so on, making doubling dilutions all the way down the row. Discard 50µl from the last well in each row.
    Positive control for haemagglutination: well F1 = 50µl 'Flu' + 50µl RBC's.

  6. Add 50µl influenza virus preparation (= 8 HAU) to all the wells - work from right to left - why?

  7. Add 50µl RBC's to all the wells - work from right to left - why?

  8. Mix the cell suspensions by tapping the plate gently.
